Abstract

Population explosion is one of the many problems faced by developing countries. Using contraceptives to limit family size is the forefront in controlling the global increment in population. But the side effects and other constraints incurred by synthetic contraceptives have called the need to use naturally occurring antifertility agents. Achyranthes aspera is an abundant indigenous herb used as an abortificent agent by people living in rural Ethiopia. Its pharmacological screening has indicated the active principles involved in its antifertility action. The present study was aimed at confirmation of the antifertility activity of Achyranthes aspera through histological and biochemical investigations on mice. 80% methanolic extract of the leaves of the plant was evaluated for its effect on the body weight, genital organ weight, folliculogenesis, endometrial cell height, stromal thickness, myometrial thickness and ovarian and uterine glycogen levels. The extract was administered to female albino mice orally by a gavage at doses of 800, 1000, and 1600mg/Kg body weight for 30 days. The control groups were treated with distilled water for the same period. In the first part of the study, both healthy and atretic follicles were counted from every tenth serially sectioned ovarian tissue, and the epithelial cell height of the endometrium, stromal and myometrial thicknesses were measured from uteri sectioned at 6µm.The ovarian and uterine glycogen levels were also quantified by the method of phenol-sulfuric acid colorimetric technique. The extract caused a significant (P<0.05) increment in the body and uterine weight, but a significant (P<0.05) decrease in the weight of ovaries in the test groups as compared to the control group. There was a significant decrease in the number of healthy ovarian follicles but a marked increment in the number of atretic follicles in the test groups than the control group. The histological observation made on the uteri showed a significant (P<0.05) increase in the endometrial cell height, uterine lumen, stromal and myometrial thickness in the experimental groups compared to the control group. The extract also caused a significant (P<0.05) increment in the glycogen level of ovaries and uteri of the test groups compared to the control group animals. In conclusion, the antifertility effect of Achyranthes aspera leaf extract on female albino mice was found to be positive which could be through inhibiting the development of recruited follicles into the next stage, creating a nonreceptive environment for the implantation of the blastocyst.
